I think you should talk to your manager one more time and get a final result from him. Then go up to the HR. I had something like this before, (I was leaving the company and wanted to take a dayoff as a floating holiday, but my mamager I cannot, because the PTO should be earned as we work there. But after I was thinking, floating holiday is nothing to do with PTO. Then I sent an email to HR and asked if I could - I did not mention my mamager said no. HR replied back and told me I can take one floating holiday out of 2 for the whole year. Then I forwarded HR's email to my manager and asked him if that would be OK with him. Guess what, he said "that will be fine")
Sometimes, you have to flight for yourself, otherwises, who will??? But do not against your manager personally. Put the attitude like you just want to clear the company's policy and make sure you are treated fairly.
