My H1-B started in Jul 2003. The last renewal was extended to Jun 2010 based on my pending I140/485.

I currently work on EAD after left my old employer in Oct 2009.
-Is it Oct. 2009 or 2008?

Can I change back to H1-B status? If yes, is it going to be a transfer or do I need to get out of the country and re-enter to activate H1-B status?
-You probably need to get an H1 visa out of the country and re-enter to activate H1-B status. But it may hurt your I-485 because your current status is not H1.

My wife is going to graduate in Dec and she needs a legal status to stay here. I filed my 140/485 before we got married and my PD is not current.
-Your wife status will be AOS after she graduates. So, she has no problem to stay and she can work using EAD.
