In this case, if I did not get REF reived email for USCIS, should I call then to ask:

1: How long should I wait before I call them to confirm.

Had better two weeks later. Also check your case status online from time to time.

2: What should I ask them when I called.

-Whatever you want to such as say you sent the response on whta date and see whether they got it or not.
