这个两年的有效期是从何时开始计算的？申请下来时？还是我正式离境的日期？

-正式离境的日期. You need to come back to US before you stay in China for two years even if the RP does not expire.

So you mean if Wang Wu applies RP on 1/1/2011 and it is approved on 3/1/2011, and he does not leave US until 4/1/2013, in this case he may be admitted on his first return on 3/31/2015?  How about he does not leave in 10 years?
