MTR should be filed to the service center where denied

your case,if you choose appeal,file I-290B & check to the service center,then write your brief to AAU (Admin. Appeal Unit) of USCIS in Washington DC.
If you file a MTR or an appeal for your I-485 & I-140,your EAD will be valid continually
