H1 and you will need H1 quota for that.But if you have US Master or Ph.D. degree, you can use the special 20000/year quota, which is much less restricted.

2. Your I140 will be affected if it is university/company sponsored. Or else, if it is NIW or EB1A, and you will be working in the similar field of your expertise (which supports your I140 petition), it will be OK.
