A job offer or permanent position is not required for EB1-A I-140. A letter of employment, demonstrating you are currently employed and doing the same / similar thing underlying i-140 petition should be needed for I-485 ( In I-140, you claim you are extraordinary in certain field of research, you may not be so in other fields. The letter will prove you are still working on your extraordinary field, but not others. In other word, the letter will show the conditions underlying I-140 petition still continues).
