Yes, you can file I-140 anytime if you believe you are eligible. The A# is assigned to you, but the I-485 you filed previously can only exist if the EB1B is not denied. If EB1B I-140 keeps pending after your NIW is approved and PD current, you can then link that I-485 to the NIW. If EB1b is denied anytime before that, I-485 is not valid and you need to refile I-485 when NIW PD is current.
