The new job is quite different, so AC21 portability is not likely to apply to your case.
Change job at the 5th year of H1 and start over a new I-140 is not a good idea, because you have only 1 year H1 left for your new job and the university needs to file for you new EB1OR+485+EAD very soon or you have to change to O-1 to work.
