Why i Can't format my h/d?

I have booted my PC from CD-ROM, and then i go to Command Prompt
type in "format C:"

but it keep telling me "Format cannot run because it is in used by other process, Format may run if this volume is dismounted first, would you like to dismount the volume first?"

I then check "yes"

but it then give me an error message saying "Cannot lock the drive, The volume is still in use".

:( pls help what should i do?
