I have a follow up for Q2, I am not exactly understand the sequence.

Would my sister family be consider as one pertition all together?

If as you said, means when my sister's priority date becomes current, suppose my nephew is in US, then my sister should not add her son into the form who will come to US with her, my nephew can only apply 1-485 again after his mom/dad becom GC holder.

Would his age become a problem?

-His CSPA age have to be < 21 at that time to file I-485. The CSPA age = the child's age when the priority date is current - the I-130 pending time.

 

Also my nephew needs to disclose his GC pertition when he apply for F1 visa right?

-Yes. His F-1 status will not affect him to file I-485 and not affect the I-485 approval.
